Kymograph

A kymograph is a 2D plot that shows particle position over time in a movie, so movement appears as lines or tracks. It is used to measure motion features such as velocity, run length, pause time, direction, and event frequency.

What it is and what it measures

A kymograph is a plot made from time-lapse movie data. One axis shows position along a chosen path, and the other axis shows time, so each moving particle can appear as a track. Researchers use it to measure motion features such as velocity, run length, pause time, direction of movement, event frequency, and in some cases particle flux and intensity-related information.

How it works

To make a kymograph, the user first traces a path in the image, often along a linear biological structure such as an axon or dendrite. The movie is then converted into a position-versus-time image. If an object moves at constant speed, its track looks like a straight line; steeper or shallower slopes reflect different velocities. More complex motion can produce curved or uneven tracks, and pauses can appear as flat segments.

Why scientists use it

Kymographs give a compact visual record of dynamic processes in living cells. They are especially useful for intracellular transport, where cargoes or motors move in anterograde or retrograde directions, reverse direction, stop briefly, or change speed. In neurons, the long, thin shape of axons and dendrites makes kymograph analysis practical for following movement along one main axis.

Limits and cautions

Kymographs are useful, but manual analysis can be slow and can vary between users. Extracting quantitative results is not always straightforward, especially in crowded images, when signal-to-noise ratio is low, or when two channels must be compared to decide whether tracks belong to the same moving object. Automated tools can help, but the underlying tracing and interpretation still matter.

SEE THE IDEA

How a kymograph represents movement

The movie is transformed into a position-versus-time plot, and the shape of each track is then interpreted as movement behavior.

A kymograph turns movie frames into a time-space record that makes motion easier to measure.
