MAPT
MAPT is a human gene that encodes tau, a microtubule-associated protein. In the sources, it is studied because changes in its regulation or mutations are linked to tau-related neurodegenerative disease.
What MAPT is
MAPT is the gene that encodes tau, a microtubule-associated protein. Tau helps with microtubules, which are internal cell structures that support cell shape and transport. In the sources, MAPT is important because altered MAPT activity is linked to tauopathies, a group of diseases marked by tau dysfunction and aggregation.

What researchers measure
The sources measure MAPT-related biology in several ways: whether DNA variants increase or decrease regulatory activity, whether promoter sequences act as cis-regulatory elements (CREs), and whether changes are specific to certain cell types such as neurons. They also measure disease-related outcomes such as tau pathology, neurofibrillary tangles, and neuroinflammation.

How MAPT is connected to disease
Tau dysregulation is described as important in progressive supranuclear palsy, frontotemporal dementia, and Alzheimer’s disease. In Alzheimer’s disease, tau can become hyperphosphorylated, meaning it carries too many phosphate groups, and then form neurofibrillary tangles, which are clumps associated with cell death. In frontotemporal dementia, MAPT mutations are reported to promote tau pathology and a reactive microglial state, meaning microglia switch into an inflammatory activation pattern.

How regulation was studied
One study screened about a 3 megabase region around the MAPT locus, which is the chromosomal neighborhood containing the gene, using massively parallel reporter assays (MPRAs). An MPRA is a lab method that tests many DNA sequences at once by linking them to a reporter readout, so researchers can compare which sequences raise or lower regulatory activity. The study found many cell-type-specific CREs and many promoter variants with increased or decreased activity in neurons.

Limitations of the evidence
The regulatory findings were generated in a limited set of cell types, mainly H1 haplotype cells, excitatory neurons, HEK293FT cells, and mixed neuron cultures. The study did not directly test how the variants change MAPT expression in living cells in a complete causal way, and the authors note that results could differ in H2 haplotype cells. The sources are preprints or preprint-based summaries, so the evidence should be treated as early rather than final.
