Messenger RNA
Messenger RNA (mRNA) is a single-stranded ribonucleic acid that carries genetic information from DNA and serves as a template for protein synthesis on the ribosome. It is usually expressed only temporarily, so the protein-making signal does not last long.
What it is
Messenger RNA, or mRNA, is a single-stranded ribonucleic acid. It is transcribed from deoxyribonucleic acid (DNA), carries genetic information, and directs protein synthesis by acting as a template on the ribosome, the cell structure that reads the code and builds a peptide chain from amino acids. Its expression is transient, meaning the message is present for only a limited time. [0,1,2]

How it works
The mRNA sequence tells the ribosome which amino acids to join and in what order. In eukaryotic cells, important parts of mRNA include a 5′ cap, untranslated regions (UTRs; segments that are not translated into protein), and a poly(A) tail. These features help control translation efficiency and mRNA stability. Longer poly(A) tails can bind more poly(A)-binding proteins, which can improve translation and slow degradation. [1,2]

Why it matters in therapeutics
mRNA can be designed quickly with bioinformatic tools and made by chemical methods, which can speed development. In therapeutics, it is often delivered by lipid nanoparticles (LNPs), but delivery is difficult because mRNA is large and negatively charged, and it can be degraded by ribonucleases in the bloodstream. The literature also reports that many mRNA products are still in preapproval clinical trials, showing that the technology is promising but still limited by stability and delivery challenges. [0,2]

Limitations
The main limitations are short half-life, susceptibility to degradation, and difficulty crossing cell membranes. Formulation components, repeated dosing, and off-target effects can also raise safety concerns. Changes to mRNA structure can improve stability and protein output, but they do not remove all delivery and toxicology issues. [0,1,2]
