Neurobasal

Neurobasal is a defined cell culture medium used to grow primary neurons in vitro, meaning outside the body in a dish. It is known for supporting neuron survival, but studies also show it can trigger unwanted neuronal stress or death under some conditions.

What it is

Neurobasal is a defined culture medium, a liquid mixture of nutrients and chemicals used to keep cells alive in the laboratory. The sources describe it as widely used for primary neuronal cell culture, especially for neurons from the brain. In this context, what is measured is how well neurons survive, how active they are, and whether they show signs of damage in vitro.

How it acts

The medium is designed to help neurons survive, but one study found that exposure to Neurobasal could kill a large fraction of postnatal hippocampal neurons after several hours, and even brief exposure could cause death measured later. The same study linked this effect to NMDA receptor-mediated excitotoxicity. NMDA stands for N-methyl-D-aspartate, a receptor type that responds to glutamate, a common brain signaling molecule. In that experiment, D-APV, a blocker of NMDA receptors, prevented the toxicity, and L-cysteine at a concentration found in commercial recipes reproduced the harmful effect.

What researchers measure with it

Scientists use Neurobasal to study neuronal survival, electrical activity, and injury responses. In one injury model, cultures maintained in Neurobasal showed more markers of pathology after stretch injury than cultures maintained in BrainPhys/SM1. Those markers included lactate dehydrogenase, or LDH, release, which is a sign of cell damage or death, and breakdown products linked to calpain activity, along with other injury-associated changes. Another result showed that synaptic stimulation increased pERK in BrainPhys/SM1 cultures but not in Neurobasal/B27 cultures, suggesting that the medium can change how strongly neurons respond to activity.

Limitations and comparison with other media

Neurobasal is useful, but it is not neutral. The sources say it may promote excitotoxicity, and its effects can differ from other media such as Minimal Essential Medium, or MEM, which did not show the same toxicity in the comparison study. BrainPhys is described as a newer medium that better supports neuronal activity and long-term viability in some settings, and it reduced injury markers compared with Neurobasal in the stretch-injury model. So results from neurons grown in Neurobasal must be interpreted with care because the medium itself can change survival and signaling.

Why this matters for mechanism

A mechanism is not just a correlation; it is an organized set of parts and activities that produces an outcome. Here, the outcome is neuronal survival, activity, or damage, and the relevant parts include the medium components, receptors such as NMDA receptors, and downstream injury processes. That means Neurobasal is not only a background fluid: it can be part of the causal setup that shapes the phenomenon being measured.

Neurobasal in neuron experiments

Neurobasal can support neurons, but in some settings its composition also changes signaling and can increase damage-related measurements.

The medium is part of the experimental condition and can influence both neuron behavior and the readouts used in the study.
