Neurotrophin / growth factor protein · BEGINNER LESSON
NT-3
NT-3, short for neurotrophin-3, is a growth factor protein that binds most strongly to the TrkC receptor. It helps neurons survive, grow axons, and develop proper electrical properties, especially in sensory systems.
What NT-3 is and what is measured
NT-3 is one member of the neurotrophin family, a group of growth factor proteins found throughout the nervous system. In studies, researchers measure where NT-3 acts and what it changes: neuron survival, axon growth or attraction, receptor binding, and electrical properties such as ion currents, membrane behavior, and action potential repolarization. Receptors are proteins on cell surfaces that receive signals; TrkC is the highest-affinity receptor for NT-3, meaning NT-3 binds it most strongly. p75NTR, the low-affinity neurotrophin receptor, can also be activated by neurotrophins. How NT-3 works
NT-3 signals through TrkC and, depending on cell context, can also interact more weakly with TrkA, TrkB, and p75NTR. This signaling can support neuron survival, promote axon outgrowth, and influence synaptic plasticity, which means changes in how strongly neurons connect and communicate. In sensory neurons, NT-3 can also act as a short-range chemoattractant, a signal that guides growing axons toward a target. In culture, blocking TrkC prevented the attraction, showing that this guidance effect depended on NT-3 signaling. [1,2]

What experiments showed about auditory neurons
In brainstem slice experiments, applying NT-3 had little effect on high-frequency neurons but strong effects on low-frequency neurons. It changed voltage-dependent potassium currents, which are ion flows that help shape how a neuron repolarizes after firing, and it also altered passive membrane properties. The effects were bidirectional during development: potassium currents decreased at one embryonic stage and increased at another, showing that NT-3 does not act the same way at all times or in all cells. The authors suggest NT-3 helps establish tonotopy, the ordered arrangement of sound-frequency processing along the auditory pathway. [0]

Limitations and scope
NT-3 is not a universal switch for every neuron. The excerpts show strong effects in some low-frequency auditory neurons and sensory axons, but minimal effects in high-frequency auditory neurons in the same study. Its action also depends on developmental stage and receptor context. The sources describe experimental findings in mice, chicks, cultured neurons, and slice preparations, so the results show biological roles, not a complete map of all NT-3 functions in every species or tissue.
