Primary cilia
Primary cilia are single, non-motile, microtubule-based structures that project from many vertebrate cells. They act as specialized sensory and signaling compartments that detect outside cues and help control cell responses.
What they are
Primary cilia are small, solitary projections on the surface of many vertebrate cells. They are built from microtubules, which are stiff protein tubes used as cell scaffolding, and they do not move fluid the way motile cilia do. Their main measured role in the sources is sensory and signaling function: they detect extracellular cues and help organize how cells respond.

Core structure
A primary cilium has three main parts: the basal body at the base, the transition zone, and the axoneme, which is the microtubule core extending outward. The axoneme usually has a 9+0 pattern, meaning nine outer microtubule doublets and no central pair. The ciliary membrane surrounds this core and forms a biochemically distinct compartment.

How signaling works
Primary cilia act like selective signaling compartments. Receptors on the ciliary surface detect ligands, downstream effectors process the information inside the cilium, and terminal effectors create cell responses. Intraflagellar transport, or IFT, moves proteins and other cargo in both directions: kinesin motors carry cargo toward the tip, and dynein motors bring cargo back toward the cell body.

Why the compartment matters
The cilium has a special gateway that controls what enters and stays inside. This selective permeability helps concentrate signaling molecules, which can increase sensitivity and reduce random noise. The sources also describe ciliary signaling as spatially separated from the rest of the cytoplasm, which helps keep some signaling events insulated from the cell body.

Limitations and dynamic behavior
Primary cilia are not permanent on every cell at every moment. They are tied to the cell cycle, often assembling in G0/G1 and disassembling later in the cycle. Their structure and signaling can also change with mechanical stress, signaling states, and protein changes such as acetylation or deacetylation of α-tubulin, a microtubule building block.

SEE THE IDEA
Primary cilium as a selective signaling compartment
Anchor the cilium to the cell and help control entry into the compartment.
Forms the structural core and supports the cilium’s shape and function.
Kinesin moves cargo outward; dynein returns cargo inward for maintenance and recycling.
Detect outside ligands and feed signals into processing inside the cilium.
Processed signals influence downstream transcription, metabolism, and other cellular outputs.
Structure and transport build a controlled compartment that turns outside signals into cell responses.
Primary cilia combine a distinctive microtubule scaffold with selective transport to organize signaling.
