Primary cilium
A primary cilium is a single, non-motile, microtubule-based projection on many vertebrate or mammalian cells. It acts as a small signaling compartment that senses and helps organize signals from outside and inside the cell.
What it is and what is measured
A primary cilium is a tiny, single projection from the cell surface made of microtubules, which are internal protein tubes that give shape and support. Scientists describe it by its presence on a cell, its non-motile nature, its ‘9+0’ microtubule pattern, and the proteins found there, such as ARL13B and AC3. These markers help identify the structure in cells and tissues. The cilium is usually measured by its length, structure, and the location of signaling molecules within it.

How it works
The primary cilium is separated from the rest of the cell by a transition zone, which acts like a gate. Proteins enter and leave by intraflagellar transport, or IFT, a delivery system that uses motor proteins to move cargo along the cilium. This transport helps build and maintain the cilium and supports signaling pathways such as Hedgehog and Wnt. Because it is a spatially restricted compartment, it can concentrate receptors and second messengers, helping the cell detect mechanical, chemical, and metabolic cues.

When it forms and what it does in cells
Primary cilia often assemble when cells are not dividing and are removed as cells re-enter the cell cycle, the sequence of growth and division. In the nervous system, neuronal primary cilia can affect axon guidance, synaptic function, and sensory transduction, which means converting a stimulus into a cellular signal. In some sensory neurons called nociceptors, they have been linked to excitability and pain signaling. In other cell types, they also influence development, metabolism, and hormone secretion.

Limitations and why the details matter
Primary cilia do not work the same way in every cell type. Their length, orientation, and protein makeup can vary by location and state of the cell. Problems in cilium formation or signaling can lead to ciliopathies, a group of disorders affecting many organs, but the exact outcome depends on which proteins are altered and whether they act inside the cilium or indirectly affect it. Some signaling links are still being worked out, so the cilium is best understood as a regulated hub rather than a simple on/off switch.
