Developmental signaling protein · BEGINNER LESSON
Sonic hedgehog
Sonic hedgehog (Shh) is a secreted signaling protein that acts as a morphogen, meaning cells read its spatial signal to adopt different developmental fates. In the vertebrate neural tube, Shh also can stimulate progenitor cell proliferation.
What it is and what is measured
Sonic hedgehog, usually abbreviated Shh, is a secreted molecule in the Hedgehog signaling pathway. In the sources, researchers measure Shh presence, its packaging into exosomes (small membrane vesicles released by cells), and the cellular responses it triggers, such as ventral neural patterning, gene-expression readouts, and progenitor proliferation. One study also follows single cells over time to compare Shh response strength with fate choice.

How Shh works in development
Shh helps pattern the ventral side of the neural tube, the early brain and spinal cord tissue. It forms an extracellular gradient, meaning its level changes with position, so nearby cells receive different amounts of signal. Cells interpret this through the Hedgehog pathway: Shh relieves inhibition of Smoothened (SMO), which then leads to downstream signaling. The main transcriptional effectors are GLI proteins, which control target genes and help cells adopt specific progenitor identities.

Two different output types
The sources show that Shh can be sorted into two biochemically distinct exosomal pools. A denser pool, Shh-P150, drives Smoothened-GLI1 signaling and supports ventral progenitor identity. A lighter pool, Shh-P450, activates a Smoothened-Gαi-dependent route that increases progenitor proliferation without pushing ventral fate. This means the same ligand can produce different developmental outcomes depending on how it is packaged and which intracellular route is used.

Mechanism and control
Rab7 is a late endosomal regulator needed for making the Shh-P150 pool and for notochord-driven ventral neural patterning. When Rab7 is lost, secretion shifts toward the proliferative Shh-P450 pool, which disrupts morphogen-patterning signals. In single-cell studies, Shh response dynamics are not perfectly uniform: cells with similar signals can still choose different fates, showing that a gene-regulatory network (GRN, a set of genes and their control links) processes Shh with some variability.

Limitations
The sources emphasize that Shh response is not read with perfect precision in every cell. Cell position, signal duration, and noise in signaling can affect fate choice, so a Shh response level does not guarantee one exact outcome in a single cell. The exosome-based mechanism is shown in the cited developmental context, so broader claims should stay within that evidence.

SEE THE IDEA
How Sonic hedgehog can lead to different developmental outcomes
Cells release Sonic hedgehog into the tissue environment.
Shh can be packaged into a dense P150 pool or a lighter P450 pool.
P150 favors SMO-GLI1 patterning; P450 favors SMO-Gαi proliferation.
Cells either adopt ventral identities or increase proliferation.
Shh packaging influences which signaling route is used, and that helps determine whether cells are patterned or encouraged to divide.
A secreted ligand can be routed into different vesicle pools, producing different cell responses.
