Streptavidin

Streptavidin is a protein that binds biotin very tightly and specifically. In biology, this binding is used to detect, localize, capture, or release tagged molecules in experiments.

What it is and what is measured

Streptavidin is measured by how strongly and specifically it binds biotin, a small vitamin. Sources describe its affinity as extraordinarily high and note that this binding is useful in experiments such as imaging, purification, and diagnostic tests. Some studies also measure how its binding changes with mirrored versions of streptavidin or biotin, or with engineered tags that can be displaced by biotin.

How it works

The main mechanism is a very strong streptavidin-biotin interaction. This lets streptavidin detect biotinylated targets, because a target can be chemically or biologically labeled with biotin and then captured or visualized with streptavidin. In one imaging method, a protein is fused to TurboID, a biotin ligase that adds biotin to nearby sites, and fluorescent streptavidin reveals where that protein is inside the cell. In another system, proteins are tagged with a streptavidin-binding peptide, trapped in streptavidin condensates, and later released by adding biotin.

Why stereospecificity matters

The sources show that streptavidin is highly stereospecific, meaning it prefers one mirror-image form of biotin over the other. Matched pairs bind far more strongly than mismatched pairs, and this difference can make the natural and mirror-image systems functionally separate. That property is being explored to reduce interference from endogenous biotin and possibly lower immunogenicity in mirror-image systems.

Limits and caveats

The sources also describe limits. Streptavidin can face endogenous biotin interference in diagnostic settings, and native streptavidin may be immunogenic, which matters for therapeutic use. In imaging, streptavidin may be better than antibodies in some cells, but signals can still differ from antibody or GFP-based readouts because protein location can change over time. Engineered condensate systems are currently best for cytosolic proteins, and biotin entry into cells can vary because biotin is cell-impermeable and depends on transport.

SEE THE IDEA

How streptavidin is used in experiments

Biotin-labeled targets are recognized by streptavidin, which creates a visible signal or an experimental handle.

Streptavidin acts like a high-affinity binder that links biotin labeling to detection or control.
