TDP-43 mislocalization

TDP-43 mislocalization means the TDP-43 protein is found in the wrong place in the cell, especially leaving the nucleus, where it normally helps process RNA. In disease studies, this shift is often linked with aggregation and reduced RNA-binding function.

What it is and what is measured

TDP-43 is an RNA/DNA-binding protein that is usually located in the nucleus, the cell compartment that holds DNA. Researchers study TDP-43 mislocalization by looking for where the protein is found, whether it forms aggregates, and whether RNA-processing or DNA repair functions change. In many studies of amyotrophic lateral sclerosis-frontotemporal dementia (ALS-FTD), TDP-43 is found outside the nucleus together with protein clumps, and this is linked to impaired cell function.

How it happens

The sources describe several linked mechanisms. TDP-43 normally helps pre-messenger RNA (pre-mRNA) splicing, messenger RNA (mRNA) transport, and the stabilization and local translation of mRNA. When TDP-43 is mutated, misfolded, chemically modified after translation, or cleaved into fragments, its RNA-binding ability can drop and the protein can become more likely to move abnormally and aggregate. One review also notes that TDP-43 normally binds damaged chromatin and supports the DNA damage response, so when TDP-43 is depleted or mislocalized, DNA repair and genome maintenance are disrupted. Some evidence also suggests a feed-forward loop: DNA damage can further promote TDP-43 mislocalisation.

Why it matters

In the reviewed studies, loss or abnormal location of TDP-43 was associated with more DNA damage, stronger DNA damage-response signaling, and poorer repair activity. For example, TDP-43-depleted cells showed many more double-strand breaks, and a mutation called Q331K reduced repair activity while increasing DNA-damage-response activation. These findings support the idea that mislocalization is not just a location change; it can be part of a chain that harms RNA handling and genome stability.

Limitations

The evidence comes from a mix of cell lines, patient-derived induced pluripotent stem cells (iPS cells), organoids, rodent models, and post-mortem tissue, so not every result comes from the same kind of experiment. The reviews also say the exact triggers for cytoplasmic mislocalization and aggregation are still incompletely understood. That means the overall pattern is strong, but some steps in the mechanism remain under study.

Supporting concepts

Nucleus: the compartment that contains DNA. RNA: a molecule made from DNA instructions that helps build and regulate proteins. Aggregation: when proteins stick together into clumps. DNA damage response (DDR): the cell’s repair-and-signal system for damaged DNA. Ribonucleoprotein particle: a complex of RNA plus protein.

TDP-43 mislocalization and downstream effects

Normal nuclear TDP-43 can be disrupted by mutation, cleavage, or other changes, leading to mislocalization, aggregation, and impaired repair.

A location change in TDP-43 can alter both RNA-related roles and DNA-repair support.
