Triton X-100

Triton X-100 (TX-100) is a widely used nonionic detergent, meaning it helps dissolve and reorganize membrane material without carrying a net charge. It is used because it can solubilize membrane proteins while often preserving their structure and function better than stronger ionic detergents.

What it is and what is measured

Triton X-100, often shortened to TX-100, is a nonionic detergent or surfactant. A surfactant is a substance that interacts with oily and watery parts of a system, and a nonionic one does this without a net electric charge. In the sources, TX-100 is measured by how it changes membranes, how well it solubilizes membranes or pollutants, and the concentration where it forms micelles, called the critical micellar concentration (CMC). Micelles are small clusters of detergent molecules in solution. The sources place the CMC around 0.2–0.3 mM and show that some membrane changes happen below that level.

How it acts on membranes

The sources describe TX-100 as first inserting into a membrane bilayer, which is a double layer of lipids that makes up a vesicle or cell membrane. At sub-CMC concentrations, this insertion can disturb lipid packing, increase fluidity, and trigger swelling without fully destroying the vesicle. One study reports a shape change from spherical vesicles to toroidal, donut-like structures, showing that the membrane can reorganize before full solubilization. A proposed three-step model is: detergent monomers saturate the membrane, mixed detergent-lipid micelles form and fragment the membrane, and then those mixed micelles are released into solution.

Where it is useful

Because TX-100 can gently solubilize membrane proteins while keeping them functional, it is widely used in biochemical and structural studies. The sources also say it is effective at solubilizing phosphocholine-rich membranes, especially in fluid phases. In environmental applications, one review describes TX-100 as a nonionic surfactant with high solubilization capacity for petroleum-based pollutants, including a combined solubilization-migration mechanism used in soil and groundwater remediation.

Limitations and cautions from the sources

The sources emphasize that TX-100 is not uniformly effective in every system. Gel-phase membranes require higher concentrations for complete solubilization, and longer lipid chains make this harder. Cholesterol can inhibit TX-100 action, likely by promoting detergent-rich regions. In soils, adsorption in organic-rich material can reduce effectiveness. The review also notes risks: TX-100 can degrade into octylphenol, which is described as endocrine-disrupting, and high concentrations can inhibit microbial activity. Another limitation is that some membrane changes happen below the CMC, so the membrane can be remodeled before full solubilization, making interpretation of experiments more complex.
