TUBB3

TUBB3 is a gene that makes one beta-tubulin protein used in microtubules, the cell fibers that help cells keep shape, move, and build axons. Changes in TUBB3 can disrupt axon guidance and brain development.

What TUBB3 is

TUBB3 is the gene for one beta-tubulin (β-tubulin) protein, called βIII-tubulin. Beta-tubulin is one part of microtubules, which are hollow protein fibers in the cell’s cytoskeleton, the internal structural framework. TUBB3 is found in nerve cells and is especially important in axons and dendrites, the long cell extensions that help neurons connect.

What it does in cells

Microtubules grow and shrink as tubulin proteins are added or removed from their ends. That behavior helps cells change shape and move. In neurons, TUBB3 helps support neurite growth, which is the growth of axons and dendrites. Microtubules also act as tracks for motor proteins, which move cargo inside cells, and TUBB3 appears important for how those motors attach to microtubules.

How it helps axon guidance

At the growth cone, the growing tip of an axon, TUBB3 connects receptor signals to microtubule dynamics. One source reports direct binding to the netrin-1 receptors DCC, DSCAM, and UNC5C in polymerized microtubules. Netrin-1 changes these contacts: it increases TUBB3 association with DCC and DSCAM during attraction and branching, and decreases TUBB3–UNC5C interaction during repulsion. Axon guidance is the process that steers axons to the correct targets.

What goes wrong in disease

Disease-causing missense mutations, which change a single amino acid in the protein, can alter microtubule dynamic instability and microtubule–kinesin interactions. Reported outcomes include axon guidance defects, cortical malformations, congenital fibrosis of the extraocular muscles, and cortical dysplasia, complex, with other brain malformations 1. In some cases, mutant TUBB3 also fails to support normal netrin-1–dependent binding patterns.

Limitations and evidence level

The sources show strong links between TUBB3 changes and altered microtubule behavior, neurite growth, and brain wiring, but they also note limits. Other beta-tubulin isotypes can compensate for basic viability, so TUBB3 is not the only beta-tubulin in cells. Some mechanisms, including how receptor–TUBB3 and motor–TUBB3 interfaces look in atomic detail, remain unresolved.
