Gene / presynaptic protein · BEGINNER LESSON
UNC13A
UNC13A is the gene that encodes Munc13-1, a presynaptic protein that prepares synaptic vesicles for fusion so neurotransmitters can be released. It is also involved in insulin granule priming and can be reduced when TDP-43 loss exposes a cryptic exon in its RNA.
What UNC13A is and what it does
UNC13A encodes Munc13-1, an essential protein at the presynaptic active zone, the site where a neuron releases chemical messenger molecules called neurotransmitters. Its main measured role is synaptic vesicle priming: it converts vesicles into a fusion-competent state so they can release neurotransmitter during evoked synaptic transmission. In glutamatergic synapses, loss of this function strongly disrupts release even when synapses still look structurally normal. Beyond neurons, Munc13-1 also primes insulin granules and is required for glucose-stimulated insulin secretion.

How it works mechanistically
Munc13-1 is controlled by several interacting domains. Its MUN domain helps promote assembly of SNARE proteins, which are the membrane-fusion machinery. The protein can also be autoinhibited by interactions among its own N-terminal C2A, CaM-binding, and acidic polyE segments with the MUN domain; this suppresses SNARE assembly. That inhibition can be relieved by RIM zinc-finger binding, calmodulin, and calcium ions (Ca2+). Ca2+/calmodulin and Ca2+/phospholipid binding also speed vesicle replenishment and affect short-term plasticity, the rapid change in synaptic strength during repeated activity.
Genetic and disease-related observations
Human UNC13A variants have been linked to different outcomes. Some coding or splice-site variants cause a neurodevelopmental syndrome through reduced expression, gain-of-function changes in neurotransmission, or altered second-messenger regulation. In amyotrophic lateral sclerosis (ALS), TDP-43 loss can expose a cryptic exon in UNC13A pre-mRNA, leading to nonsense-mediated decay, a quality-control process that destroys RNA with premature stop signals. This lowers UNC13A protein levels and helps connect TDP-43 pathology with synaptic failure. Population studies also report UNC13A haplotype blocks that influence disease severity and survival, but these associations can be population-specific and need replication.

Important limits
The sources show strong functional evidence for vesicle priming and regulation, but not every interaction has the same level of proof. For example, some population associations are statistical and do not by themselves prove causation. Also, the clinical studies cited here describe associations in specific cohorts, so the results may not transfer to all populations without further validation.
