Whole-cell capacitance

Whole-cell capacitance is the membrane capacitance measured after a patch-clamp electrode breaks into a cell and makes the cytoplasm electrically continuous with the pipette solution. It reflects how much cell membrane is present and is used as an indirect sign of cell size and sometimes cell health or cell type.

What it is and what is measured

Whole-cell capacitance is the membrane capacitance, written as Cm or Cp in the sources, obtained after the whole-cell patch-clamp configuration is established. Capacitance is the ability of a membrane to separate and store electrical charge. In this setup, the plasma membrane acts like an insulator between the solutions inside and outside the cell, so the measured capacitance depends on the cell membrane surface area. Larger neuronal cells generally have higher capacitance, and the value can help distinguish cell type or indicate cell health in context.

How it arises in whole-cell patch-clamp

Patch-clamp uses a glass micropipette to form a tight seal, called a gigaseal, with the membrane. When the membrane patch under the tip is ruptured, the pipette solution becomes electrically continuous with the cytoplasm, allowing the amplifier to measure whole-cell currents and membrane properties. At that point, the recording system can estimate capacitance together with series resistance, also called access resistance. The measured capacitance is not a separate structure; it is the electrical result of the cell membrane separating charge across the membrane surface.

Why it matters in experiments

Capacitance is useful because it gives an indirect estimate of membrane surface and can support interpretation of recordings from different cell types. The sources also note that changes in capacitance can appear when moving from cell-attached to whole-cell recording, and that the value can be used alongside series resistance to judge the quality of the recording. However, the measured value depends on the configuration and the recording conditions, not just on biology alone.

Limitations and caveats

Whole-cell recordings are invasive. After break-in, the cell interior can be dialyzed, meaning some intracellular molecules can diffuse into the pipette, and that can disrupt molecular machinery needed for some phenomena. The sources also note that whole-cell measurements do not localize currents to small subcellular regions, and that series resistance can distort the true membrane voltage during voltage-clamp recordings. So capacitance is helpful, but it should be interpreted together with other recording limits.

SEE THE IDEA

How whole-cell capacitance is obtained

Seal first, break-in second, then measure capacitance and interpret it with the recording context.

Whole-cell capacitance is measured only after access to the cell interior is established.
